After opting to skip the 3-0 one-day international series whitewash, Gayle will again sit out both games at the Sir Vivian Richards Cricket Ground in Antigua.
A media release from the West Indies Cricket Board confirmed the big-hitting opener had asked to miss the series, which begins on Saturday, although it is expected he will be back for the two Tests later this month.
While Gayle will be absent the hosts welcome back skipper Darren Sammy, after he was also rested in the ODIs, in a 13-man squad that includes eight members from last year's World Twenty20 final success.
Leg-spinner Samuel Badree was one of the revelations of that tournament and the 31-year-old school teacher will look to further his reputation after impressing in the recent Caribbean T20.
"I'm looking forward with great expectations to the matches ahead," he told the WICB website. "We had a good one-day series against the Zimbabweans where we won all three matches, and we will also look to win both Twenty20 matches."
Zimbabwe have lost their past 14 Twenty20 internationals against Test-playing nations. Their most recent win was, however, against West Indies in Trinidad three years ago.
The tourists are hopeful in-form batsman Craig Ervine will be able to return after an asthma attack forced him the miss the final ODI.
Team physiotherapist Amato Machikicho told Zimbabwe Cricket's website they remained cautious.
"We want a gradual return that does not strain him so after he pulled through the batting session, I asked the coach to excuse him from the fielding," Machikicho said.
