GeneMaker Gene Synthesis is available for less than the cost of
PCR cloning. According to Blue Heron Bio, their novel, non-PCR
GeneMaker gene synthesis provides perfectly accurate clones rapidly and
economically.
 
GeneMaker’s proprietary software designs the build strategy and
stores the instructions in a database. Double-stranded oligonucleotides
(“duplexes”) are made to the specifications designated by the design
software by GeneMaker’s proprietary gene assembly instrument. The gene
assembly instrument sequentially adds “duplexes” to a solid matrix in a
proprietary column.
 
Blue Heron claims three advantages of gene synthesis over PCR: 1)
this process can be used to assemble any length DNA fragment, from 50
base pairs to 40,000 base pairs or longer; 2) the sequential addition
avoids errors of order that commonly occur during PCR synthesis, and 3)
the automation enables high throughput, highly accurate gene production
quickly and at a low cost.
